Introduction
Mitosis is the nuclear division associated with the asexual division of cells. In multicellular diploid organisms, mitosis takes place during the division of somatic cells. Mitosis distributes chromosomes equally to each of daughter cells ensuring that every cell in an organism carries the same set of chromosomes. Mitotic activity is the foundation for the development and growth of organism. Two main division activities of living cells are karyokinesis and cytokinesis. Karyokinesis is the process of partitioning genetic material into daughter cells during nuclear division, which is complex and requires great precision. The two daughter nuclei produced will each have a chromosome composition identical to that of the parent cell. Cytoplasmic division or cytokinesis follows karyokinesis. Cytoplasma material is partitioned into two parts, and then encloses both new cells within a distinct plasma membrane. Cytoplasmic organelles are replicated or synthesized anew in each cell.
Objectives
Prepare and stain slides of onion root tips.
Observe cells in the process of mitosis.
Form and test a hypothesis about the effects of fertilizer on mitosis.
Determine the effect of fertilizer on the percentage of cells in mitosis.
Materials
• Aceto-orcein stain in dropping bottle (30 mL)
• Lab apron
• Coverslips
• Compound light microscope
• Distilled water
• Eyedropper
• Gloves
• 1 M HCl in dropping bottle (6 mL)
• Microscope slides
• Onion root tips, specially grown
• Paper
• Paper towels
• Glass watch
• Mounting needle
• Forceps
Procedure
1. Write a hypothesis that clearly states how you think the concentration of fertilizer affects mitosis in onion roots.
2. Put on safety goggles, gloves, and a lab apron.
3. Use forceps to carefully remove three onion root tips from the vial labeled “Y—100%,” and place them in a glass watch.
4. Use an eyedropper to flood the root tips with 1 M HCl. Allow the root tips to stand in the HCl for 10 minutes. CAUTION: HCl is an irritant and a poi- son. Avoid skin and eye contact. If you get HCl on your skin or cloth- ing, wash it off at the sink while notifying your teacher. If you get HCl in your eyes, promptly flush it out at the eyewash station while notifying your teacher. Notify your teacher in the event of an acid spill.
5. Use the eyedropper to remove the HCl from the glass watch, and dispose of it as instructed by your teacher. Be careful to not remove the root tips with the eyedropper. Refill the glass watch with distilled water.
6. Place a microscope slide on a paper towel. Add three drops of aceto-orcein stain to the center of the slide. CAUTION: Aceto-orcein stain is corrosive and poisonous. Avoid eye and skin contact. In case of ...
